Melanocytes from normal healthy adult controls and from the nonlesional epidermis of one patient with vitiligo (skin type IV) were grown in Hams F-10 medium with 10% fetal calf serum. 12-O-tetradecanoyl phorbol-13-acetate, and cholera toxin. All mitogens were removed from the media 7 days before the harvest of these cells. All control cultures were melanotic, but the vitiliginous cells had low cell density and decreased pigmentation.

    • -3 M EDTA at 37°C for 6 hours. The incubation was stopped by acidic iodine oxidation with 1% iodine in 2% KI in HCL (1 M) for 1 hour. The excess of iodine was removed with 1 M ascorbic acid followed by centrifugation and separation with the use of ion-pair reversed-phase HPLC. Specific activity was determined by quantitation of neopterin and its di- and triphosphate produced from GTP.
