14. -3 M together with the removal of growth factors. Subcultures of the second or third passage were used in the experiments reported herein. Melanocytes from normal healthy adult controls and from the nonlesional epidermis of one patient with vitiligo (skin type IV) were grown in Hams F-10 medium with 10% fetal calf serum. 12-O-tetradecanoyl phorbol-13-acetate, and cholera toxin. All mitogens were removed from the media 7 days before the harvest of these cells. All control cultures were melanotic, but the vitiliginous cells had low cell density and decreased pigmentation.

16. -3 M EDTA at 37°C for 6 hours. The incubation was stopped by acidic iodine oxidation with 1% iodine in 2% KI in HCL (1 M) for 1 hour. The excess of iodine was removed with 1 M ascorbic acid followed by centrifugation and separation with the use of ion-pair reversed-phase HPLC. Specific activity was determined by quantitation of neopterin and its di- and triphosphate produced from GTP.
