Penetrances for ovarian cancer in carriers of the breast cancer susceptibility gene were derived from E. B. Claus, J. M. Schildkraut, W. D. Thompson, and N. J. Risch [Am. J. Hum. Genet. 53, A787 (1993)], which estimated a cumulative frequency of 10% by age 60. The effects of age were incorporated into the analyses as in (2). Male breast cancer cases were as to the same liability classes as female breast cancer cases diagnosed below age 30. All cancers other than breast and ovarian cancer were ignored in this analysis Allele frequencies were those reported in the Genome Data Base and were based on 56 CEPH (Centre d'Etude du Polymorphisme Humain) chromosomes.
