    • Morphology and Phenotype of Dendritic Cells from Peripheral Blood and Productive and Non-productive Infection with Human Immunodeficiency Virus Type 1
    • of outstanding interest, Dendritic cells from 24 hour cultures of peripheral blood mononuclear cells were identified from morphology and phenotype using immunogold labelling and electron microscopy. A series of cell types probably reflecting different stages of life-history was identified with small numbers of cells having blunter pseudopods (reminiscent of LCs) and a few with a very veiled/interdigitating cell type of morphology. Productive infection with HIV-1 was only seen with these two possibly more mature morphologies.

    • Primary Prolifera tive and Cytotoxic Responses to HIV Induced in Vitro by Human Dendritic Cells
    • of outstanding interest, Primary stimulation with antigen-pulsed DCs using cells from donors not infected with HIV-1 and who are not, therefore, immunocompromised were used to identify a small peptide that is a T-cell epitope. In individuals of different MHC types, it initiated production of cytotoxic T cells that killed virus-infected target cells. This approach shows the efficiency of DCs in primary stimulation and should identify peptides with protective potential for vaccination purposes.

    • Cells that Present Both Ligand and Costimulatory Activity Are the Most Efficient inducers of Clonal Expansion of Normal CD4 T Cells
    • of outstanding interest, Delivery of a ligand plus a co-stimulatory molecule, two neceassary signals for clonal expansion of T cells, was achieved either on a single cell or on two separate cells. Delivery of both signals on one cell appeared to be at least 30 times more efficient than separate delivery of the signals. The authors speculate that this could explain why tissues can express autoantigens and contain antigen-presenting cells without inducing autoimmunity.
