Their 11-residue linker would then be only eight residues in length, and their addition of one or two (GGGGS) segments may have increased activity simply by increasing linker length.

    • This group found that deleting only two FR residues caused nearly a 50-fold increase of binding affinity as well as a significant change in specificity. Observation of this pronounced effect on hapten binding suggests the possibility that, in some situations, fusion of a linker segment between the pair of V domains of a particular Fv could have an effect on binding site properties in the resulting sFv analog.
