    • Fetuses were obtained by cesarian section from anesthetized, timed-pregnant cats with sterile surgical technique (1). After intracardial perfusion with 4% paraformaldehyde in 0.1M sodium phosphate buffer, the telencephalic vesicles were exposed and crystals of DiI were inserted with a fine needle. A total of eight cortical injections were made in five fetuses at E30. The brains were stored in the same fixative at room temperature for 1 to 4 months, depending on the age of the fetus, to allow for diffusion of the dye. Coronal sections were cut on a vibratome at 100 to 200 μm and stored in fixative for subsequent viewing with rhodamine epifluorescence optics. Sections were counterstained with meta-phenylene diamine which is visible with fluorescein optics

    • DiI was placed in the dorsal thalamus of fixed cat brains at several fetal ages: E30 (two fetuses), E36 (two), E43 (three), E51 (three), E60 (three), and P7 (two). Because the brains were fixed, extremely precise DiI injections could be made, even into discrete thalamic nuclei such as the lateral geniculate nucleus in the older (E43 and on) fetuses. In addition to the cat fetuses, two P14 ferrets received DiI injections bilaterally into the SC. Because of the immaturity of ferrets at birth, the cortex of a P14 ferret is comparable to that of an E5 8 cat [(1)

    • After diffusion of the dye, 100- to 200-μm coronal or horizontal sections of cat or ferret brain were cut on a vibratome. In many sections a photooxidation reaction was carried out to convert the fluorescent label into a stable, electron-dense reaction product. DiI-labeled sections were incubated in a cold, filtered solution of diaminobenzidine (DAB; 75 mg in 50 ml of tris buffer, pH 8.2) for 2 hours before being illuminated under rhodamine epifluorescence optics. A dense DAB reaction product on Dil-labeled cells is clearly visible at the end of the photooxidation reaction. This procedure is similar to ones published independently
