In carboxypeptldase solution a tyrosyl residue coordinates to Zn2+ i.e., is much closer to the metal ion than in the crystals. The high enzymatic activity toward carboxyamide substrates found for Co2+-, Ni2+-, and Zn2+-carboxypeptidase A, 112 in which the metal ion has a five-coordinate, 112 octahedral, 112 and distorted tetrahedral108 coordination sphere, respectively, may also suggest a flexible active site. The metal ion In carboxypeptidase A also imparts substrate specificity; it discriminates between esters and peptides. The authors113 conclude that the results observed “would be expected If the metal atom were to interact with the carboxyl group of esters during the binding step but with the carbonyl group of peptides during the catalytic step”.
