An evaluation of the hypoxic regulation of three ocular cell types, including endothelial cells, pericytes, and pigment epithelial cells. Hypoxia induced VEGF expression in all cells in a dose-dependent and reversible manner. Endothelial cell growth was stimulated by exogenous VEGF and by media conditioned under hypoxic conditions by ocular cells. VEGF-neutralizing compounds eliminated both of these responses, suggesting VEGF was the primary hypoxia-induced mitogen under these conditions.

Proc Natl Acad Sci U S A 1995, 92:905-909. This study demonstrated that VEGF expression was correlated both temporally and spatially with the onset of ischemia-induced retinal neovascularization in the mouse. Protein localization predominated at Muller-like cells, but mRNA expression was primarily found in the inner nuclear retinal layers.

Hypoxic regulation of VEGF expression in the intact developing retina was demonstrated by using oxygen-enriched atmospheres that inhibited vessel formation and endogenous VEGF production.

The first indirect evidence that VEGF may play a role in diabetic retinal hyperpermeability; VEGF protein expression and serum albumin extravasation were colocalized using immunohistochemistry in diabetic rats. As compared with control retinas, VEGF expression was increased in diabetic retinas.

Evaluation of 210 intraocular fluid samples from 164 patients (143 from diabetic individuals) demonstrated that both aqueous and vitreous VEGF concentrations were high during active intraocular neovascularization and low at other times. The predicted intraocular concentration gradient and VEGF reduction following successful treatment with laser photocoagulation were also demonstrated.

Proc Natl Acad Sci U S A 1995, 92:10457-10461. Injections of VEGF-neutralizing receptor chimeric proteins were given intravitreally in a murine model of ischemia-induced retinal neovascularization. Retinal neovascularization was suppressed in 95% to 100% of animals evaluated. The inhibition was dose dependent and occurred without detectable toxicity by light microscopic evaluation.
