J Clin Endocrinol Metab 1995, 80:1347-1351. Includes a comprehensive study of the TSHR gene, gsp, and ras gene mutations in 37 thyroid hyperfunctioning adenomas. The study is unique because all three genes are examined in each sample. Mutations were not found simultaneously in any sample, suggesting that each gene can play an independent role in the development of hyperfunctioning thyroid adenomas.

Rarity of oncogenic mutations in the thyrotropin receptor of autonomously functioning thyroid nodules in Japan

Takeshita A, Nagayama Y, Yokoyama N, Ishikawa N, Ito K, Yamashita T, Obara T, Murakami Y, Kuma K, Takamatsu J, Ohsawa N, Nagataki S: Rarity of oncogenic mutations in the thyrotropin receptor of autonomously functioning thyroid nodules in Japan. J Clin Endocrinol Metab 1995, 80:2607-2611. Shows a low frequency of TSHR gene mutations in hyperfunctioning thyroid adenomas in Japan. One possible explanation of this finding may be a geographic variation because Japan is a relatively iodine-sufficient area compared with Europe.

2+ cascades

2+ cascades. Mol Endocrinol 1995, 9:725-733. This is the first study in which the entirety of exon 10 of the TSHR gene was examined in 11 hyperfunctioning thyroid adenomas. Activating mutations have been identified in about 80% of toxic adenomas. In two cases the researchers were able to constitutively activate both the regulatory cascades normally under the control of TSH.

The cyclic adenosine 3′,5′-monophosphate-responsive factor CREB is constitutively activated in human somatotroph adenomas
-
Bertherat J, Chanson P, Montminy M: The cyclic adenosine 3′,5′-monophosphate-responsive factor CREB is constitutively activated in human somatotroph adenomas. Mol Endocrinol 1995, 9:777-783. Reports the presence of activated CREB protein in a series of pituitary adenomas activated independently by the presence of a gsp mutation. This suggests the involvement, in the gsp-negative samples, of other elements of the cAMP cascade. Because of the close analogy between thyroid and pituitary hyperfunctioning tumors, the same hypothesis can be postulated for thyroid pathologies.

Russo D, Arturi F, Schlumberger M, Caillou B, Monier R, Filetti S, Suarez HG: Activating mutations of the TSH receptor in differentiated thyroid carcinomas. Oncogene 1995, 11:1907-1911. Provides the first evidence of activating mutations of the TSHR gene in differentiated thyroid carcinomas. The mutations were found in a series of carcinomas selected for their constitutive high adenlyl cyclase activity. In one tumor the simultaneous presence of a TSHR gene mutation and N-ras mutation has been identified.
