This study demonstrated that animals that do not carry a functional copy of the insulin-sensitive glucose transporter were still able to maintain relatively normal glucose homeostasis. This rather surprising result indicated the presence of compensatory glucose transport systems.

    • Hotamisligil GS, Murray DL, Choy LN, Spiegelman BM: TNF-α inhibits signaling from insulin receptor. Proc Natl Acad Sci U S A 1994, 91:4854-4858. This paper describes the mechanism by which TNF-α induces insulin resistance in cultured adipocytes and demonstrates the ability of TNF-α to reduce insulin-stimulated tyrosine phosphorylation of the insulin receptor and IRS-1. In addition, this study also demonstrated that in cultured cells other cytokines can interfere with insulin receptor signaling.

J Clin Invest 1994, 94:1543-1549. This paper demonstrates that TNF-α is, at least in part, responsible for the reduced tyrosine kinase activity in obesity in whole animals. After neutralization of TNF-α, a significant increase was observed in the signaling capacity of the insulin receptor in obese and insulin resistant fa/fa rats.

    • Increased adipose expression of tumor necrosis factor-α in human obesity and insulin resistance
    • Hotamisligil GS, Arner P, Caro JF, Atkinson RL, Spiegelman BM: Increased adipose expression of tumor necrosis factor-α in human obesity and insulin resistance. J Clin Invest 1995, 95:2409-2415. These authors describe the overexpression of TNF-α in adipose tissue of obese humans. They also demonstrate the corrective decreases in adipose TNF-α expression after a weight loss program that results in improved insulin sensitivity.

    • Improvement in glucose tolerance and insulin resistance in obese subjects treated with troglitazone
    • Nolan JJ, Ludvik B, Beerdsen P, Joyce M, Loefsky J: Improvement in glucose tolerance and insulin resistance in obese subjects treated with troglitazone. N Engl J Med 1994, 331:1188-1193. In this work the clinical importance of the treatment of insulin resistance in obesity is demonstrated. Furthermore, the agent used in this study illustrated the fact that thiazolidinedione compounds are effective in the treatment of insulin resistance in humans.
