This paper shows that it takes only a low dose of supplemental dehydroepiandrosterone to return serum levels in older individuals to those of younger controls, but this supplementation has surprisingly few dramatic effects on metabolic parameters of aging. However, it has a dramatic effect on the hard- to-define sense of well-being. A clue that the central nervous system may be the most pertinent site to search for the site of action for dehydroepiandrosterone.

Only when the molecular mechanism of action of dehydroepiandrosterone is known will it be possible to understand how it interacts with glucocorticoid hormones. Although dehydroepiandrosterone may not work through currently envisioned mechanisms that involve the glucocorticoid receptor, this paper reviews much of the evidence that at least some of its actions involve antagonizing the effects of glucocorticoids.
