    • The high-level stable expression in Chinese hamster kidney of a G-protein-coupled seven-helix receptor is achieved. For gene amplification, a single plasmid with the dehydrofolate reductase gene under the control of the simian virus 40 promoter and the receptor gene under the control of the strong cytomegalovirus immediate early promoter is transformed into the cells and clones are selected in the presence of increasing methotrexate concentrations. The inclusion of a receptor antagonist is necessary to stabilize the receptor.
