Wierzbicki A, Sikes CS, Madura JD, Drake B: Atomic force microscopy and molecular modelling of protein and peptide binding to calcite. Calcif Tissue Int 1994, 54:133-141. An intriguing attempt to use atomic force microscopy and energy minimization calculations to study a biologically relevant interaction. It is shown that poly-L-aspartic acid molecules bind in parallel fashion to the (001) face of calcite crystals. An aspartate-rich protein, from mollusk shell is also observed to be non-randomly oriented on (110) 'terrace' faces of calcite. Molecular modelling showed that the (001) face is the preferred site for interaction of poly-L-aspartic acid in the β-pleated sheet conformation. Favourable interactions also occur with the (110) face, both parallel and perpendicular to the c-axis. The predicted binding to the (110) and (104) faces is much weaker.

An attempt to obtain structural information about a class of proteins strongly implicated in calcium carbonate nucleation. RP-1, a protein from the soluble matrix of scallop shell, is shown to contain an internal sequence of eight contiguous aspartic acid residues. RP-1 is a potent inhibitor of calcium carbonate crystallization but, interestingly, has no effect on calcium phosphate formation.

This study illustrates the variety of ways in which organic matrix molecules can influence the growth habit of calcite. Whereas glycoproteins from sea urchin spine induces the formation of additional (10/) faces in calcite, glycopeptides derived from these proteins induced the formation of (401) faces, and deglycosylated protein induces the formation of (203) faces.

FASEB J 1995, 9:262-268. The latest in a valuable series of studies using high-energy synchrotron radiation to examine the effects of occluded proteins on echinoderm skeletal elements. By binding to specific planes of the calcite crystals, these proteins decrease the size and increase the misalignment of the coherence domains, so greatly reducing the tendency of these crystals to fracture along the (104) cleavage planes. Effects of occluded proteins on the shape of the coherence domain may also influence the growth habit of the skeletal element.

This study describes a novel protein that is a major component of chicken egg shell. Ovocleidin-17 is a 17 kDa protein which is rich in hydrophobic and basic amino acids, and is highly aggregated in aqueous solution. Using immunocytochemistry, it is shown that ovocleidin is present throughout the palisade layer and outer shell membrane.

J Struct Biol 1995, 115:50-59. Electron microscopy of developing enamel reveals the presence of chains of electron-lucent spheres oriented perpendicular to the dentin-enamel junction. These spheres appear to be similar to those formed in-vitro by recombinant amelogenin. The authors propose that the amelogenin chains promote enamel crystal growth along the c-axis and determine intracrystalline spacing.

The first demonstration of a means by which the organic matrix of a mineralized tissue may determine the formation of a specific crystal polymorph. Soluble proteins from various shell types are combined with β-chitin and silk fibroin to produce a matrix similar to that of mollusk nacre. Under these conditions, soluble proteins from calcite-containing structures induce the formation of calcite, whereas proteins from aragonite-containing structures induce the formation of aragonite.

2+ ions are appropriately spaced to interact electrostatically with the carboxylate groups of the (β-pleated sheet) polypeptide. In contrast, phosphorylated proteins (phosphophoryn and phosvitin) bind to the (010) faces, apparently by substituting their phosphorylated side-chains for the phosphate groups of the crystal.

A synthetic oligonucleotide designed to cleave amelogenin mRNA is shown to transiently inhibit production of amelogenin protein in mouse enamel. This treatment results in severe hypomineralization, characterized by a complete absence of hydroxyapatite prisms in molar inner enamel.
