Gardella TJ, Luck MD, Jensen GS, Usdin TB, Jüppher H: Converting parathyroid hormone-related peptide (PTHrP) into a potent PTH-2 receptor agonist J Biol Chem 1996, 271:19888-19893. The PTH-2 receptor is activated by PTH, but only poorly or not at all by PTHrP. The article shows that the substitution of a single amino acid residue, trypophan instead of phenylalanine at position 23, dramatically enhances the binding affinity of PTHrP for the PTH-2 receptor; replacement of histidine by isoleucine at position 5 turns PTHrP into a full agonist at the PTH-2 receptor.
