The nucleotide sequence of the entire nonviral insert of the ASV 16 genome and of the flanking viral DNA was determined in both strands with the Sequenase version 2.0 kit (U.S. Biochemical).

39. The pBS-v-P3k 19 plasmid was digested with Sac I and Apa I. The v-P3k-Sac I-Apa I fragment (minus the env gene) was cloned via an adaptor plasmid into the avian retroviral vector RCAS (5). Replication in CEFs yielded RCAS-v-PSk. The cellular clone pBS-c-P3k was digested with Sac I and Aat II. The c-P3k-Sac I-Aat II fragment was similarly cloned via an adaptor plasmid into RCAS. This plasmid was designated as RCAS-c-PSk. DNA transfection and focus assay were carried out as described (21).
