The problem of lateral confinement in fluorescence microscopic studies was recently analyzed: J. Hendricks, T. Kawakatsu, K. Kawasaki, and W. Zimmermann, Phys. Rev. E 51, 2658 (1995). Moreover, if one of the two degrees of freedom of a polymer in three-dimensional space is projected out by observing only a two-dimensional image of the contour, the corresponding variables are no longer independent [J. Wilhelm (private communication)]. The interpretation of microrheological data is complicated by the fact that the method is neither microscopic nor macroscopic in a strict sense.
