Proc Natl Acad Sci USA 1997, 94:6831-6836. This paper describes a novel method, selective amplification via biotin- and restriction-mediated enrichment (SABRE), for detection of differentially expressed mRNAs. This procedure uses selective streptavidin-biotin affinity and restriction enzyme site reconstitution to enrich for cDNA species more abundant in one population than in another.

This paper reports the application of the cDNA microarray technology to identify novel human genes. In these experiments, microarrays containing 1046 human cDNAs of unknown sequence were printed on glass with high-speed robotics and the resulting DNA 'chips' were used to quantitatively monitor differential expression of the cognate human genes using a highly sensitive two-color hybridization assay. Array elements that displayed differential expression patterns under given experimental conditions were characterized by sequencing.
